effect of listeria monocytogenes on tumor immunotherapy with dendritic cells

چکیده

introduction: different microbial compounds can induece maturation of dendritic cells (dcs) into dc1 or dc2 .in the present study, we used tumor antigen pulsed dendritic cell that matured with listeria monocytogenes antigen and cholera toxin (ct) in the treatment of a murin model of cancer. material and methods: tumor cells (wehi-164 balb/c derived fibrosarsoma) were injected subcutaneously to balb/c mice. bone marrow cells were cultured with gm-csf and il-4.on day 5, listeria monocytogen lysate or ct) cholera toxin (added to culture for 2 day. for immunization, 106 dendritic cells matured with ct or listeria monocytogenes (l.m injected) subcutaneously around the tumor site. results: we found that immunotherapy with dendritic cells treated with listeria antigens leads to delay in tumor growth as compared to the ct and control groups. dcs treated with listeria produce higher amount of il-12 copmare to ct and control groups. conclusion: some microbial components such as listeria monocytogenes that favors the immune response towards th1 could be used in generating powerful dcs for cancer immunotherapy.

Listeria Monocytogenes Activated Dendritic Cell Based Vaccine for Prevention of Experimental Tumor in Mice

Background: The use of dendritic cells (DCs) as a cellular adjuvant provides a promis-ing approach in immunotherapy of cancer. It has been demonstrated that Listeria mono-cytogenes activated DCs pulsed ex vivo with tumor antigens trigger a systemic Th1-biased specific immune response and a single dose of this vaccine will cause a consider-able anti tumor immunity. Objective: The present study w...

Dendritic Cell Maturation with CpG for Tumor Immunotherapy

Background: Bacterial DNA has immunostimulatory effects on different types of immune cells such as dendritic cells (DCs). Application of DCs as a cellular adjuvant represents a promising approach in the immunotherapy of infectious disease and cancers. Interaction of Listeria monocytogenes with mouse dendritic cells.

In this study, the interaction of murine dendritic cells with Listeria monocytogenes was investigated. Dendritic cells are efficient antigen-presenting cells, play a key role in the immune response, and are capable of migrating over substantial distances between sites of infection and lymphoid tissues. L. monocytogenes EGD invaded dendritic cells, escaped from phagosomes into the cytoplasm, and...

Dendritic cells: emerging roles in tumor immunotherapy.

The purpose of this article is to examine the use of myeloid dendritic cells (DCs) as immunotherapy in the treatment of cancer. DCs can be stimulated either from circulating blood or bone marrow progenitor cells using cytokines, particularly granulocyte macrophage-colony-stimulating factor (GM-CSF) (e.g., sargramostim, Leukine). GM-CSF has been shown to promote maturation, mobilization, and ant...
